Zongertinib: Breakthrough ERBB2 Lung Cancer Treatment!

[00:00:00]This is pretty darn good, actually, for something that was hard to target before. So, ERBB2 or HER2 mutations are like EGFR mutations. Um and, you know, this ERBB2 mutations often happen in exon 20, which was that untargetable region of EGFR.

[00:00:17]So, the first strategy that was actually used was this antibody drug conjugate called trastuzumab deruxtecan, antibody that's been used in breast cancer since like 1998.

[00:00:27]And then deruxtecan, old chemotherapy, linked together to bring it into the cells. Response rate's high, around 50%.

[00:00:36]Yeah, time to progression, yeah, 10 months plus.

[00:00:40]But now we're getting pills that actually work on this. So, zongertinib is a pill that was rationally designed to try and get into that mutant form of the protein, that hard to target ERBB2 exon 20. And and now response rate's high, 70%ish, yeah, progression-free survival over a year. This is pretty darn good, actually, for something that was hard to target before. Um but two new FDA FDA-approved options within the last year. And zongertinib just got first-line FDA approval, also, for this type of lung cancer in the metastatic setting.

[00:01:13]Um so, zongertinib's now an option, and trastuzumab deruxtecan, zongertinib, afatinib um are also effective, too.
