Chromatography is [music] a separation technique that separates substances based on their different affinities towards two phases, the stationary phase and the mobile phase.
>> [music] >> There are several types of chromatography, including paper chromatography, thin layer chromatography [music] or TLC, gas chromatography, and liquid chromatography. [music] In our investigation today, we will use paper chromatography to analyze lipstick [music] pigments.
Now, let's discuss why lipstick analysis is important. Lipstick contains different substances such as dyes, pigments, oils, and waxes.
Analysis of lipstick is important for several reasons.
First, it helps manufacturers maintain product quality and ensure consistent color. Second, [music] it can help identify counterfeit cosmetic products.
Third, it can be used in forensic investigation [music] to compare lipstick samples found at a scene. Finally, it helps ensure that cosmetic products are safe [music] for consumers by detecting harmful substances.
Okay, what is principle chromatography?
>> [music] >> Principle of chromatography is based on the separation. Different pigments dissolve in differently [music] in the solvent and therefore they travel at different speeds along the chromatography paper.
In this experiment, [music] the chromatography paper is acts as the stationary phase while the solvent acts as the [music] mobile phase.
As separation occurs, different colored bands are formed on the paper. [music] >> Lipstick chromatography involving two phases, which is stationary [music] phase and mobile phase. In studying lipstick chromatography, the stationary phase is the chromatography [music] paper which remain fixed while sample is processed.
While mobile phase is the solvent that [music] travels through the stationary phase carrying the dissolved pigment with.
In advanced analytical settings, scientists [music] may use uh specialized extraction techniques such as magnetic solid phase extraction to isolate specific lipstick components before [music] they are analyzed through the column.
>> To perform this analysis, we use lipstick samples, chromatography paper, ethanol or [music] acetone as the solvent, a capillary tube, beaker, pencil, and ruler. [music] These materials allow us to carry out paper chromatography efficiently and safely.
>> First, [music] a baseline is drawn on the chromatography paper using a pencil.
The lipstick sample [music] is then applied onto the line using a capillary tube.
Next, solvent is poured into a beaker and the chromatography [music] paper is carefully placed inside ensuring that the sample spot remains above the solvent level.
>> As the solvent moves [music] upward through capillary action, the pigments separate. Finally, [music] the paper is removed, dried, and the separated pigment bands are observed.
>> [music] >> Let's calculate our RF value. Our RF value is determined by [music] dividing the distance traveled by the pigment by the distance traveled by the solvent front.
For example, [music] if a pigment travels 4.0 cm while the solvent front travels 8.0 cm, [music] the RF value is 0.5.
Different pigments [music] have a different RF value, which helps in their identification.
>> The result of lipstick chromatography is [music] a visual chromatogram featuring distinct color bands that serve as a unique chemical fingerprint of the sample. By analyzing these bands, researchers can identify [music] specific red colorants, detect hazardous contaminants like heavy [music] metals, and uncover illegal additives such as sulfonamide antibiotics.
