The Glycocalyx: The largest organ you've never heard ofAdded:
If I asked you to name the largest organ in your body, you'd probably say the skin. But, there's a far larger organ located deep inside your thousands of miles of blood vessels. It's a massive nano infrastructure of sugars and proteins called the glycocalyx, and it functions as a highly intelligent filter for everything entering the cells. To really understand its scope, consider that the glycocalyx and its broader integrated structure, the endothelial surface layer, a gel-like matrix filling the space between the blood and the cell wall, can often be more than 300 times thicker than a cell's phospholipid membrane. As just one of its many functions, the glycocalyx is a critical regulator of oxygen delivery because while the well-known transporter protein hemoglobin carries oxygen, the glycocalyx and the endothelial surface layer ensure this oxygen reaches tissues through optimizing blood flow physics and signaling for vessel dilation. What this really means is that through ensuring the red blood cells are evenly distributed across capillary networks, the glycocalyx maximizes the total surface area available for hemoglobin to offload oxygen, while the endothelial surface layer acts as both a lubricant and a buffer against the considerable friction created by the rush of plasma and red blood cells. Essentially, the glycocalyx acts like seaweed in the tide. The delicate fibers of the glycocalyx bend as blood flows over them, and this bending regulates blood pressure through telling the vessel walls beneath to relax, widen, or tighten up.
The health and vitality of the glycocalyx is also a significant predictor of cardiovascular health as atherosclerosis, or the progressive accumulation of plaque in the arteries, really begins with the thinning and shedding of the glycocalyx. Because when this protective layer is damaged, cholesterol and inflammatory cells can finally reach and adhere to your vessel walls. The glycocalyx is extremely sensitive to an inflammatory diet as even a single high-sugar meal or any spike in blood sugar can cause the glycocalyx to shed to the point of near collapse. While the chronically high blood sugar seen in diabetes keeps the glycocalyx consistently thinned. This is a large reason behind the array of commonly experienced diabetic vascular complications, and also one major reason why diabetics are more susceptible to infections and heart disease. The deterioration and dysfunction of the glycocalyx is a virtually universal feature of the aging process, unfortunately, and it's driven, again, by a systemic thinning of the glycocalyx that does allows inflammatory cells and proteins to reach the vessel wall more easily.
So, how do we maintain the health and vitality of our largest organ as we age?
The most direct way is really through regular exercise, which obviously increases the heart rate and the sheer stress of blood flow, which then activates a master transcription factor called Kruppel-like factor 2, or KLF2, which forces the cell to produce more hyaluronic acid, which is the structural backbone of the glycocalyx. KLF2 forces the upregulation of the enzyme hyaluronan synthase 2, the primary metabolic enzyme that builds the glycocalyx through pushing fresh high molecular weight hyaluronic acid out to the vessel walls to repair the shedding.
In addition to regular exercise, you can also support production of the hyaluronan synthase 2 enzyme through regularly taking supplemental hyaluronic acid, which acts as a signaling molecule that directs fibroblasts to increase their production of endogenous hyaluronic acid. Vitamin D, especially as its magnesium-activated steroid form calcitriol, is an essential part of regenerating the glycocalyx and reducing the cytokine-driven chronic inflammation that degrades it. Vitamin C neutralizes excessive oxidative stress while also recycling tetrahydrobiopterin, a critical cofactor for nitric oxide production. And this is important because healthy nitric oxide levels ordinarily keep the blood vessels dilated and the glycocalyx intact. Also, original flushing niacin, or nicotinic acid, is already well-known for its unparalleled support of the tunica intima, or the innermost layer of the blood vessels. And niacin's restorative benefits extend also to the glycocalyx because the intima is also the structural foundation for the glycocalyx, and niacin supports glycocalyx longevity most by decreasing both nitrotyrosine and NADPH oxidase, two inflammatory markers of oxidative damage that generally increase with age and are especially corrosive to the delicate glycocalyx.
I hope this gives you some deeper insight into your internal mechanics, and particularly helps you understand that the massive nano infrastructure we call the glycocalyx is as much a bodily organ as the liver, lungs, skin, and pancreas, and that minimizing the age-related shedding of the glycocalyx is really a first line of defense against heart disease, the chronic low-grade assault of inflammaging, and the bidirectional reciprocal actions of these ugly and uncomfortable conditions.
